Methylene blue has been around longer than most research chemicals — Heinrich Caro first synthesized it at BASF back in 1876. What keeps it relevant nearly 150 years later is a combination of unique redox-cycling properties and the ability to plug directly into mitochondrial electron transport. Whether you’re running bioenergetics experiments, looking at oxidative stress pathways, or studying neuroprotective mechanisms, methylene blue gives you a well-characterized starting point backed by decades of published data.
Loti Labs offers research-grade methylene blue capsules at 10mg per capsule, verified above 98% purity by third-party HPLC. For any experiment where compound quality directly affects reproducibility, that analytical verification matters.
Methylene blue is a phenothiazinium heterocycle — three fused rings with a central thiazine, dimethylamino groups at positions 3 and 7, and that distinctive deep blue color in its oxidized state. The key to its usefulness is that it flips between two redox forms: oxidized MB⁺ (blue) and reduced leucomethylene blue MBH (colorless). That reversibility is what makes it work as an electron shuttle in mitochondrial research.
The molecule is planar enough to cross biological membranes easily and accumulates in mitochondria, which is exactly where you want it for organelle-level redox studies. Because it absorbs strongly at 665 nm, you can track its redox state spectrophotometrically in real time during experiments.
At its core, methylene blue is an alternative electron carrier. In the oxidized form, it picks up electrons from NADH — either through Complex I or directly in the mitochondrial matrix — and the resulting leucomethylene blue hands those electrons off to cytochrome c. This lets the cell maintain membrane potential and keep making ATP even when Complex I or Complex III aren’t working properly. For anyone studying mitochondrial dysfunction, that bypass mechanism is the main reason methylene blue is in the toolkit.
Beyond electron transport, methylene blue targets several other molecular pathways:
Methylene blue is a standard reference compound in mitochondrial bioenergetics. It consistently increases oxygen consumption rates and restores membrane potential in cells treated with Complex I or III inhibitors, commonly used in Seahorse XF and Clark electrode experiments.
Neuroprotection research has gained momentum, with studies showing methylene blue reduces oxidative stress markers, preserves mitochondrial function under stress, and modulates autophagy. It inhibits paired helical filament formation at micromolar concentrations in vitro, making it valuable in Alzheimer’s disease research.
In redox biology, methylene blue serves both as a probe for cellular redox status and as an active modulator. Its ability to cycle between oxidized and reduced forms allows study of electron transfer kinetics and NADH/NAD⁺ ratios.
Its absorption in the 600–700 nm range and efficient singlet oxygen generation make it practical for photodynamic antimicrobial inactivation against bacteria, fungi, and viruses.
All Loti Labs products, including methylene blue capsules, are for research use only. They are not dietary supplements, therapeutics, or for human or veterinary use. This designation ensures regulatory compliance and supports legitimate scientific research.
